Work Assignment Overview:

To review the literature on the current status of methodologies used to predict the pharmacokinetics (PK) of compounds metabolised by Aldehyde Oxidase (AO) and the associated issues and challenges surrounding such predictions. To use this knowledge to develop a system that will allow the accurate prediction of the human PK for in-house test compounds metabolised by AO to allow preclinical observations to be put into context of clinical relevance

Work Assignment Goals:

  • To critically review the literature on AO and provide an up-to-date summary document of the current status and challenges associated with predicting the PK of compounds metabolised by AO
  • To collate key PK and in vitro data from the literature for AO marker compounds and build a reference excel database summarising this information
  • To use knowledge of the issues and challenges associated with prediction of PK for AO compounds, to identify the best in vitro system to do this
  • To develop lab skills to allow the generation of key in vitro data needed to develop an in vitro in vivo extrapolation (IVIVE) system to accurately predict the human PK of AO marker compounds in-house. To generate a database of this information
  • To extrapolate this system to in-house test compounds that are metabolised by AO to allow confident prediction of their human PK and subsequent provision of a go/no go path forward for such molecules

Skills/Educational Background Required:

  • Ability to search literature and extract key data
  • Excellent excel skills
  • Competent in the lab
  • Ability to learn quickly and pick up key skills to work relatively independently

Skills to be Learned:

  • Key DMPK in vitro and analytical lab techniques
  • In depth knowledge of the drug metabolising enzyme Aldehyde Oxidase and the challenges associated with predicting PK for compounds metabolised by AO
  • Understanding and learning about the data required for human PK prediction and how this is done
  • Expand knowledge on how to search the literature for data and extract key information required for IVIVE
  • Improve excel skills
